1.4. Title : Integrated project Biocompact: Built-in, compact system for the handling and conditioning in sterile
environment of injectable drugs derived from innovative biotech research and from chemical-physical
molecular studies and analyses
ABSTRACT:
Goal of the project is the development of innovative solutions to maintaining the chemical-physical characteristics
of biomolecules in the entire packaging process that must take place in a controlled, aseptic
environment, as per injectable liquid drug production standards.
DESCRIPTION:
Main project goals:
In 2001 the European Council invited the Commission to examine measures aimed at developing biotechnologies
and bridging the gap between Europe and more advanced countries in the sector. Biotechnological products derive
from high-cost research run by specialist small-medium size labs. The standards of the relevant bodies are extremely
strict in granting authorisation for the use of new molecules as drugs. Quickly approving new biodrugs, created by
research, will encourage the development of small- and medium-size companies in the sector. Complying with and
maintaining the chemical-physical characteristics of biomolecules is therefore vital and the entire packaging process
must take place in a controlled, aseptic environment, as per injectable liquid drug production standards.
The packaging system aimed at handling these products must meet the following requirements:
- Compact size, easy to transport, elimination of line set-up times since the built-in module, already complete with
utilities on board the machine, will be supplied already validated and may even be installed in small spaces
- Low purchase price and running costs (savings on electricity, water, etc.)
- Adoption of ?disposable? dosing systems to avoid cross contamination of products
- Adoption of a single SW to ensure repeatability of container washing/sterilisation, product distribution, container
closure processes
- Rapid operational interventions guided by interactive system
- Reduction of environmental impact for disposal of refluent (elimination of wash detergents)
- Safety for operators (elimination of chemical sterilisation products). The knowledge acquired in the biotech sector,
with the search for new materials that do not yield particles which can be used for conditioning,
may be adopted to increase the quality of systems dedicated to the traditional pharmaceutical sector.
Contents
Content INFO SHEET N°1 : Design and manufacture of a compact module for the packaging of injectable liquids
derived from Biotechnological Research. The main characteristics of the new module will include: compact
size, low cost and quick set-up times. The main prerogative is to be shipped ready assembled in all its parts
including the utilities, after the production process having been validated during the final inspection
at I.M.A. The management SW, able to support all the more innovative and efficient liquid filling systems, allows
rapid installation. Compared with a traditional packaging line, the module can be made operational much more
quickly, allowing biotech labs to produce the first batches of new drugs for on-site experimentation in a very short
time.
Content INFO SHEET N°2 : Indication of the chemical-physical characteristics of biotechnological molecules for
analysing their compatibility with the distribution systems that can be used for packaging them in sterile
environment.
Experimental
verification
of
the
absence
of
alterations
in
the
structure
of the molecular chain of the drug after packaging. Study of new molecules, where applicable, for the creation of
innovative drugs able to combat some genetic alterations more effectively.
2
Content INFO SHEET N°3: Study and development of SW specific for the integrated module able to manage the
entire production process: container washing/sterilisation, liquid product distribution, vial closure with checking of
single operative phases of the entire biotech compact. All solutions aimed at guaranteeing repeatability of processes
must be adopted in order to allow the validation of the operating system in compliance with CFR 21-part.11. The
management of liquid product distribution must be compatible with all filling systems that may be adopted on the
compact for packaging products in compliance with injectable drug production standards (sterile environment, etc.).
Other activities compatible with the research programme.
Content INFO SHEET N°4: Study and development of innovative “disposable” filling systems for the handling of
Biotech liquids to avoid cross-contamination of products to be distributed on the packaging machine. The system
must ensure compliance with injectable liquid drug production standards (product sterility, dosing precision, etc.)
and allow repeatability of the process in order to obtain validation.
Content INFO SHEET N°5: Study and development of new biodegradable materials in the ?disposable? filling
system components to increase the safety of the handled product, to facilitate the disposal, after use, of disassembled
equipment to safeguard the environment. Certification and traceability of materials used for components.
Content INFO SHEET N° 6: Consultancy related to the development and co-ordination of an exhaustive research
which considers deep activities of data mining and gives scientific valency and measurable contents of: tests and
deepenings about problems and chances showed by the field of the research on biopharmceuticals with regard to the
required plants for the production of injectable biopharmceuticals; the identification of the necessary strategies to
the fulfilment of plants for distribution process; the evaluation of market impact of community policy for research of
policies for sustainable development, in order to plan the R&D activities linked to such strategies.
Content INFO SHEET N° 7: Consultancy for the definition of the project specifications: the starting point is what
obtained at the end of the research stage on the state of the art. The partner will have the assignment to individualize
the characteristics of the product in order to verify its full correspondence to the objectives, qualitative and
quantitative, defined by the coordinator of the project: particularly, the electronic academic researcher will define
the analysis of the software, holding in debt account both the technical-constructive aspects that the maintenance
requirements and those due to the management of the format change;
Content INFO SHEET N° 8: Consultancy related to the choice of new materials for all the parts which are in
contact with biotechnological products; it’s a matter of guaranteeing the maintenance of the purity of
the products, having a reference for simple and economical solutions, based on an easy maintenance and
substitution. The steel and, as a rule, the metals are not generally used , except considering and testing the
single sectors of employment case by chance: our research will be based on the of non-conventional materials and
their surface modification for packaging sector and on their possible application.
Content INFO SHEET N° 9: Check and development of validation procedures for entire biotech drug packaging
process
Content INFO SHEET N° 10 : Consultancy for the definition of the project specifications: the starting point is what
obtained at the and of the research stage on the state of the art. The partner will have the
assignment to individualize the characteristics of the product in order to verify its full correspondence to the
objectives, qualitative and quantitative, defined by the project: particularly, the academic researcher in mechanics
will deal him with to be of assistance for the choice of the devices and their correct sizing.
Content INFO SHEET N° 11: Consultancy related to the use of the “laminar flow”, or to the study, sizing and test of
the treatment of the air dedicated devices; will be needed a careful study of the movements and components
geometry crossed by the laminar flow, with the objective to eliminate the air points turbulence and stagnation. The
solutions to be adopted will have to be simple and economical, giving a particular attention to the energetic saving
and the environmental impact, with the purpose to minimize them. The search will also based on correlated devices
(pomps, filters, etc) study and audit and, particularly, on their performance to high temperature (300/350 °Cs and
over) and when operating in ISOTECNIA or in RABS Restricted Access Barrier System controlled areas.
Content INFO SHEET N° 12: Consultancy concerning a complete final test of the prototype and control of the
correct working of all the devices which compose it: in particular the correspondence will have to be verified to
the specifications established, identifying and signalling him the possible anomalies or the differences with respect
to the original specifications.
Content INFO SHEET N° 13: Consultancy concerning the final operating verification, to take place in a context
identical to that expected in the project seat (research laboratory): the partner will have the task in depth to try the
machine, using it in the packaging of the greater number of categories and typologies of biotecnological products.
Content INFO SHEET N° 14: Dissimination of the research results through seminars, worskshops, meeting with
industries and other interested researchers in universiy congress centre.
