STABILITY OF URINARY LOW MOLECULAR WEIGHT PROTEIN a decreased level of α1M in few native urine samples when stored at 4°C for 30 days or frozen, stored and then thawed. Degradation of α1M observed in our experiment, can be probably attributed to the procedure of freezing and thawing per se of urine samples [38]. In the study of Tencer et al. [38], degradation of α1M was related to the specificity of urine samples and the level of metallo‑proteinases. These effects, however, can be prevented by the buffer composed with inhibitors of serine‑ and metallo‑proteinases and an antimicrobial agent [40]. In addition, several studies involving individuals with various types of renal disorders suggest that α1M might be a less sensitive marker of proximal tubular dysfunction than RBP and probably also less specific in that α1M is more often elevated in both tubular and glomerular diseases [29,37,41]. For instance, in patients with burn injury-induced tubular damage, Yu et al. [41] reported weaker correlations between α1M and β2M or RBP than between β2M and RBP. The three proteins usually followed the same pattern of increase, but the magnitude of changes in RBP and β2M was two to three times greater than that of α1M. For these reasons, urinary α1M could be more useful for screening of tubular or mixed proteinuria rather than for a very sensitive and specific assessment of the proximal tubule function integrity. The stability of proteins may be influenced by several parameters, which may be different for different sets of urine samples [42,43]. For example, protein in high urinary concentrations may not be affected by various storage conditions, whereas this may not be true for low concentrations [38]. The positive effect of creatinine concentration in urine samples was found only for β2M stability. Thus indirect confirming thesis of Herber [43] that creatinine concentration of 0.7 g/L can be regarded as the threshold level for deciding whether or not urine can be analysed for β2M. The presented study confirms that α1M is the most resistant protein in acid urine, however, acid ranges of pH in the native urine are found only occasionally; by contrast, this protein was more sensitive to longer storage in alkaline buffer and to freezing and thawing. Thus, concerning at least the type of urine samples (pH) in the present investigation, the following conclusions seem to be rational and of practical value: concentrations of β2M, RBP and α1M are stable in native urine stored for 24 h at pH 6.0 or pH 9.0; storage of alkalised native urine for © Copyright by the Polish Society of Toxicology, 2005 Original Papers 91 T. Ocenę wydalania z moczem β2-microglobuliny (β2M), białka wiążącego retinol (RBP) i α1-microglobuliny (α1M) powszechnie stosuje się do określenia sprawności funkcji kanalików proksymalnych nerek. W praktyce laboratoryjnej ważnym problemem wiarygodności testów jest sposób obchodzenia się z różnymi próbami moczu i ich przechowywania. Przeprowadzono serię badań, definiując zakres zmienności i mechanizm nietrwałości białek β2M, RBP i α1M w moczu patologicznym. Szacowano, jaki wpływ na stężenie białek w próbach moczu mają zmiany pH, przechowywanie w różnych temperaturach oraz cykle zamrażania i odmrażania. Stężenia β2M, RBP i α1M w moczu oznaczano z zastosowaniem metody lateksowo-immunologicznej. Wyniki badań potwierdzają, że z ww. białek najbardziej odporna na kwaśny odczyn moczu jest α1‑microglobulina. Stwierdzono też, że α1‑microglobulina jest mniej odporna na długie przechowywanie w moczu buforowanym zasadowo, oraz jest wrażliwa na cykle zamrażania i odmrażania. W uogólniającym wnioskowaniu dotyczącym wyników różnych badań nad stabilnością białek w moczu należy zachować ostrożność. β2‑microglobulina degradowała się w patologicznym moczu przy pH 6,0 w 37°C oraz przy pH 5,5 w temperaturze pokojowej, ale na jej stabilność miały wpływ własności moczu pacjentów. Na stabilność białek w moczu może wpływać wiele parametrów i mogą być one różne dla różnych zestawów prób moczu. Otrzymane wyniki sugerują, że najbardziej prawdopodobnym powodem obserwowanej zmienności jest degradacja enzymatyczna. Ponieważ pacjenci różnią się stopniem wydalania różnych enzymów, badający powinni uwzględniać zmiany stabilności niskocząsteczkowych białek w moczu pacjentów i wcześniej zdefiniować warunki gromadzenia moczu do dalszych badań.
